Abstract: Around 90% of clinical drug trials fail due to lack of efficacy. As an important first step in the drug discovery process, improvements to the identification of promising drug target candidates could lead to dramatic gains overall. I will discuss some of the work my group has been doing on machine learning for prediction of therapeutic targets, for instance based on the concept of synthetic lethality. I will also address challenges we are finding along the way and lessons learned, involving data biases and the need to look beyond performance metrics.
