Cancer is known as one of the leading cause of death in the world with difficult diagnose at early stages, poor prognosis and high mortality. Animal-based experiments and clinical trials have always been the main approach for cancer research, albeit they may have limitations and ethical issues. Mathematical modeling as an efficient method is used to predict results, optimize experimental design and reduce animal use. Our work focuses on the phenomenological simulation of cancer progression and therapies at the cell scale level.
We develop several cell-based models to simulate pancreatic cancer progression, its interaction with immune response and the drug-oriented therapy at early stages. Meanwhile, a cellular automata model is developed to mimic cancer growth in a large scale and its recession under virotherapy. Moreover, a deformable model is set up to phenomenologically show the shape evolution of one cell and its nucleus during cancer metastasis. Furthermore, Monte Carlo simulations are performed to quantitatively investigate the uncertainties and influence of the input parameters. The corresponding mathematical methods underlying the modeling are given in details in the thesis. In summary, our mathematical models are able to help the understanding of biomedical processes during cancer progression and predict the likelihood of therapy cure.
