I am working on the reconstitution of a septin-actin cortex in GUVs.

My project focuses on the reconstitution of a septin cortex inside biomimetic lipid membranes. Specifically, I encapsulate septin, a filamentous cytoskeletal protein, in GUVs. I investigate septin localization on the inner leaflet of the membrane and its effect on the membrane mechanics.

I investigate cancer cell migration and mechanics under confinement using microfluidic devices for single cells and clusters. With this project, we hope to find a correlation between the mechanics/ deform-ability of the cells and their migration mode(s). This might lead to a better understanding of how cancer cells metastasize.

Most structural properties in (human) connective tissues are provided by collagen and hyaluronic acid networks, which occupy the extracellular matrix. These composite networks span many material properties, yet the underlying mechanisms remain poorly understood. In this project, I will investigate how such a composite network reacts to applied stress and strain by visualizing the fibers in the material during variable applied forces.

I will research how cells affect their extracellular matrix by considering cells as physical objects by incorporating microgels in extracellular matrix materials as cell mimics. Examples of such extracellular matrix materials are collagen or hyaluronic acid. I aim to observe how the “extracellular matrix” structure changes due to the microgels.
